Can this tow hook handle being used with a highlight jack to lift the vehicle?
No, these are not rated for vertical lifting.

Can this rust? And, what is the actual diameter of the hole?
Yes this can rust, it is not finished. It is actually designed to be installed only when you need it. Otherwise stored in spare and safety kit. The inner diameter is 1-1/2"
